Characterization of monoclonal antibody against human ADAM15. Lysates of Escherichia coli transfected with the expression vector FLAG-ADAM15 containing a cDNA fragment encoding
the cysteine-rich domain of human ADAM15 (lanes 1, 3, 5 and 6) or the pFLAG-CTC vector
alone (lanes 2 and 4) were immunoblotted with anti-FLAG antibody (lanes 1 and 2) or
anti-ADAM15 antibody (240-2C7) (lanes 3-6) as described in Materials and methods.
The absorption study was carried out by incubation of the antibody with 1000-fold
excess amounts of the peptide for 16 h at 4°C (lane 6). The arrow indicates the protein
band of the cysteine-rich domain reactive with anti-ADAM15 antibody. Note that no
staining is present with mock transfectants (lanes 2 and 4), and that the immunoreactive
band with anti-ADAM15 antibody (lane 5) completely disappears after reaction with
the antibody absorbed with the peptide (lane 6).
