The interleukin-1-beta (IL-1β)-induced upregulation of lipocalin-type prostaglandin
D synthase (L-PGDS) mRNA expression requires de novo protein synthesis. Chondrocytes were incubated with cycloheximide (CHX) (10 μg/mL) for 30 minutes prior
to stimulation with 100 pg/mL IL-1β for 48 hours. Total RNA was isolated and reverse-transcribed
into cDNA, and L-PGDS mRNA was quantified using real-time polymerase chain reaction.
Results are expressed as fold changes, considering 1 as the value of untreated cells,
and represent the mean ± standard error of the mean of four independent experiments.
*P < 0.05 compared with cells treated with IL-1β alone.
